Lee's agar is used specifically for differential enumeration of bacteria especially starter bacteria that grown in yogurt. Both Staphylococcus thermophillus and Lactophillus acidophillus are symbiotic bacteria that essential. 

Lee agar has a specific proportion of constituents that prevent the growth of Lactophillus acidophillus initially. S. thermophillus can ferment both sucrose and lactose. However, L. acidophillus can ferment lactose but it ferments sucrose at a very slow rate.
